Mechanism of Action

  • D2 receptor agonist
    • in anterior pituitary stimulates inhibitory dopamine receptors
    • inhibits prolactin  secretion
  • effects on 5-HT and adrenoceptors

Pharmacokinetics

  • administered orally
  • 90% absorbed through small intestine
  • metabolised in liver
  • excreted via bile
  • half-life 6-8 hours

Side-Effects

Low dose

  • constipation
  • nausea
  • postural hypotension

High dose

  • nasal congestion
  • dry mouth
  • metallic taste
  • cramps in legs
  • dystonic reactions
  • visual hallucinations
  • cardiac arrythmias
