Lamotrigine


Indications

  • Used in seizures when other drugs have not been effective
  • Used either as a sole treatment or as an adjunct treatment with other anti-epileptic drugs

Mechanism of action and pharmacology

  • Inhibits transmitter release (e.g. glutamate) by prolonging the inactivated state of the sodium channel
  • Care required in choosing the initial dose and subsequent titration
  • Valproate increases plasma-lamotrigine concentration whereas the enzyme inducing antiepileptics reduce it

Contra-indications

  • Contraindicated in hepatic and renal impairment
