Used 

Mechanism of Action

  • Close K+(ATP)
    • &rarr depolarization and Ca2+ influx
    • &rarr vesicular fusion and secretion and increased insulin synthesis
  • Risk of hypoglycaemia directly related to duration of action
    1. Short-acting e.g. Gliclazide, Tolbutamide (2x day)
    2. Long-acting e.g. Glibenclamide (1x day)

Side-effects

  • wt gain
  •  hypoglycaemia
  • GI disturbance
  • headache

Contraindications

  •  porphyria

Caution

  • Breastfeeding
  • elderly
  • hepatic / renal impairment
