Tamoxifen


Indications

  • hormonal treatment of choice in women with oestrogen-receptor positive Breast Cancer
  • delays growth of metastases
  • increases survival

Mechanism of Action

  • induce Gn release by occupying oestrogen receptors in the hypothalamus
  • this interferes with feedback mechanisms for FSH and LH release

Cautions

  • occasional cystic ovarian swellings in pre-menopausal women
  • occasional hypercalcaemia if bony metastases
  • increased risk of thromboembolic events when used with cytotoxics
  • porphyria

Contra-indications

  • pregnancy
  • breast feeding

Route

  • oral

Side-Effects

  • Hot flushes
  • vaginal bleeding or suppression of vaginal bleeding in some pre-menopausal women
  • vaginal discharge
  • pruritus vulvae
  • GI disturbance
  • light headedness
  • tumour flare
  • falls in platelet counts
