Lamotrigine
Indications
- Used in seizures when other drugs have not been effective
- Used either as a sole treatment or as an adjunct treatment with other
anti-epileptic drugs
Mechanism of action and pharmacology
- Inhibits transmitter release (e.g. glutamate) by prolonging the
inactivated state of the sodium channel
- Care required in choosing the initial dose and subsequent titration
- Valproate increases plasma-lamotrigine concentration whereas the enzyme
inducing antiepileptics reduce it
Contra-indications
- Contraindicated in hepatic and renal impairment
Adverse effects
- rashes
- flu-like symptoms
- visual problems
- dizziness
- drowsiness
- GI problems
- aggression
