Tamoxifen
Indications
- hormonal treatment of choice in women with oestrogen-receptor positive Breast
Cancer
- delays growth of metastases
- increases survival
Mechanism of Action
- induce Gn release by occupying oestrogen receptors in the hypothalamus
- this interferes with feedback mechanisms for FSH
and LH release
Cautions
- occasional cystic ovarian swellings in pre-menopausal women
- occasional hypercalcaemia if bony metastases
- increased risk of thromboembolic events when used with cytotoxics
- porphyria

- Hot flushes
- vaginal bleeding or suppression of vaginal bleeding in some pre-menopausal
women
- vaginal discharge
- pruritus vulvae
- GI disturbance
- light headedness
- tumour flare
- falls in platelet counts
